DERC - University of Colorado Vector Core :

The Vector Core is the newest core in the Barbara Davis Center Diabetes and Endocrinology Research Center. The Vector Core currently offers adenovirus production services. In the future, the Vector Core will provide more services including other viral productions (Lenti), cell lines, vectors, etc.

 

Services :

Adenovirus Production: Vector Core utilizes Invitrogen The ViraPower TM Adenoviral Expression system, which facilitates highly efficient delivery of target gene to dividing and non-dividing mammalian cells using replication incompetent adenovirus. The full Adenoviral package includes:

  • Recombination of the gene of interest from an entry clone into an adenoviral expression clone.
    Production of adenovirus in 293A cell-line, plaque purification and amplification of the adenovirus.
    Testing the expression with western blot analysis.
    Large scale preparation of the virus followed by 2 x CsCl purification and dialysis to storage buffer, OD reading and testing the stock is active.
    Quantity testing; titering.
    Quality testing; supernatant rescue assay and PCR.

Additional services:

  • MOI/time course study available.
    The cloning of the gene of interest into the entry vector available at DERC MBCF.
